Sciele Pharma And Plethora Solutions Holdings PLC Announce Initiation Of Pivotal Phase III Trials For PSD502 For Premature Ejaculation

Sciele Pharma, Inc. ("Sciele") (NASDAQ:SCRX), a specialty pharmaceutical company, and Plethora Solutions Holdings PLC ("Plethora") (AIM:PLE), the specialist developer of products for the treatment and management of urological disorders, jointly announced that they have initiated patient enrollment for the Phase III clinical trials in the U.S. for PSD502 for the treatment of premature ejaculation. In May 2007, Sciele signed an exclusive licensing agreement for PSD502 with Plethora Solutions.
PSD502 is a proprietary formulation of two marketed drugs - lidocaine and prilocaine - dispensed by a metered dose aerosol and developed for the treatment of premature ejaculation, a disorder affecting up to 30% of men in Europe and the U.S. and for which there currently is no FDA-approved prescription treatment. The product is applied locally when needed (often referred to as "on demand" therapy) and is effective within five minutes. The product has been shown to increase ejaculation latency time substantially when compared with placebo in a multi-center Phase II study. This data was published recently in BJU International (Dinsmore W et al 2007, BJU Int 99: 369-35).

Study Details:
Plethora intends to run the two pivotal studies in parallel, one in the U.S. and one in Europe. Each will be a multi-center, randomized, double-blind, placebo-controlled efficacy study recruiting a total of 540 patients across both studies. Patients will be treated for a 12-week period with an optional five-month open-label phase. generic lexapro online buy The primary endpoints will be Intra-vaginal Ejaculation Latency Time (IVELT), sexual satisfaction and ejaculatory control. Secondary endpoints will include Sexual Quality of Life and partner satisfaction. The study design and powering, in agreement with both U.S. and E.U. regulatory authorities, employ validated patient reported outcomes (PROs). The two studies will recruit patients in 36 centers in Europe and 38 in the U.S. and Canada, including almost all North American opinion leaders. A recent study shows that men who watched sexy videos or handled lingerie sought immediate gratification-even when they were making decisions about money, soda, and candy.
Authors Bram Van den Bergh, Siegfried DeWitte, and Luk Warlop (KULeuven, Belgium) found that the desire for immediate rewards increased in men who touched bras, looked at pictures of beautiful women, or watched video clips of young women in bikinis running through a park.
"It seems that sexual appetite causes a greater urgency to consume anything rewarding," the authors suggest. Thus, the activation of sexual desire appears to spill over into other brain systems involved in reward-seeking behaviors, even the cognitive desire for money.
"After they touched a bra, men are more likely to be content with a smaller immediate monetary reward," writes Bram Van den Bergh, one of the study’s authors. "Prior exposure to sexy stimuli may influence the choice between chocolate cake or fruit for dessert."
The authors believe the stimuli bring men’s minds to the present as opposed to the future. "The study demonstrates that bikinis cause a shift in time preference: Men live in the here and now when they glance at pictures featuring women in lingerie. That is, men will choose the immediately available rewards and seek immediate gratification after sex cue exposure."
Do all straight men respond the same? Actually, no. Some men are highly responsive to rewards while others are not so sensitive, and the more reward-sensitive men are the impatient ones.

Discovery of protein that helps predict prostate cancer survival

Oregon Health & Science University Cancer Institute researcher has identified a protein that is a strong indicator of survival for men with advanced prostate cancer. The C-reactive protein, also known as CRP, is a special type of protein produced by the liver that is elevated in the presence of inflammation.
"This could mean that a simple blood test that is already available could help in clinical decision making and patient counseling. Beer’s research will be published online in the journal Cancer on Monday, April 21.
Past research has shown that cancer causes an inflammatory response. This research also suggests that inflammation may play an important role in driving prostate cancer progression and resistance to therapy. Inflammatory cells are attracted to cancer sites and this local inflammation can lead to a release of inflammatory markers, like CRP.
"While inflammation may sometimes slow the progression of the cancer, an increasing body of evidence suggests that cancer can actually take advantage of the inflammatory response, and the reaction of the immune system may fuel cancer progression. To the extent that our hypothesis proves true, C-reactive protein may be reflecting the overall intensity of the inflammation," Beer said.
The finding that higher CRP is associated with shorter survival and a lower probability of response to chemotherapy is a result of a secondary analysis of inflammatory markers in patients enrolled in the ASCENT study, a large Phase 2 clinical trial that evaluated treatment with docetaxel and DN-101, a high dose formulation of calcitriol or docetaxel with placebo. This analysis included patients from both groups. The analyses were supported by Novacea Inc., the sponsor of the ASCENT study. This new finding was in collaboration with Novacea.
Because this is the first time CRP has been linked with both response and survival in study subjects with advanced prostate cancer receiving chemotherapy, it will be important to confirm this finding in an independent data set before this can become a routine blood test for men with advanced prostate cancer, Beer explained.
"If confirmed, besides providing useful information for the patient, this finding could also provide us with vital insight into the fundamental role of inflammation in the progression of advanced prostate cancer. A better understanding of this process could provide us with novel therapeutic interventions for control of this disease and its symptoms," Beer said.
